Disclosed are an oncolytic virus formulation and a preparation method thereof. The oncolytic virus formulation comprises cell vesicles derived from apoptotic tumour cells and oncolytic viruses which are coated in the cell vesicles and used as an effective constituent. The oncolytic virus formulation uses the cell vesicles derived from the cell itself to coat the oncolytic viruses therein, so as to avoid attacks by the body's immune system, and can be targeted to the treated tumour treatment site, and the anti-tumour effect is improved.
