Triglycerides determination - by saponifying oxidizing to formaldehyde, and reacting with ammonia and acetylacetone

Triglycerides in e.g. serum are determined by a procedure in which glycerol is released from the triglycerides by saponification and then is oxidised to HCHO, and the resulting HCHO is reacted with NH3 and a stable, water- and alcohol-soluble, colourless metal complex (pref. an alkali(ne eart) complex) of acetylacetone so as to give 3,5-diacetyl-1,4-dihydrolutidine (I). Pref. the reagent soln. which is reacted with the HCHO is a 1-10% soln. of a metal complex of acetylacetone in a 5-30% aq. soln. of an ammonium salt. Use of acetylacetone in complexed form gives a stabilised reagent which is still sufficiently reactive to give the desired coloured cpd. (I) with NH3 and HCHO.
