<p>PURPOSE: To obtain a new hybridoma giving an antibody capable of specifically measuring thymosin ah in high sensitivity by fusing a myeloma cell with a thymosinαl- resistant cell and subsequently selecting a strain producing an antibody recognizing the N-terminal of the thymosinαl. CONSTITUTION: The hybridoma producing a monoclonal antibody resistant to the thymosinαl is obtained by chemically modifying synthetic thymosinαl, reacting the modified thymosin with bovine serum albumen (BSA), mixing the prepared thymosinαl-BSA conjugate with an adjuvant, administering the mixture into a BALB/c mouse, repeating supplemental immunizing treatments, collecting the pancreatic cells of the finally immunized mouse, fusing the collected cells with myeloma cells, selectively culturing the fused cells in a HAT culture, screening the selectively cultured product to select the strain producing the objective antibody, cloning the selected strain by a limiting dilution method, etc., subjecting the cloned strains to the epitope analysis of the produced antibody, and subsequently selecting a hybridoma producing the monocional antibody recognizing the N-terminal of the structure of the thymosinαl. The obtained objective thymosinαl monocional antibody-producing hybridoma is useful for the diagnosis of thymoma, immunodeficient diseases, autoimmune diseases, etc.</p> |
