The recombinant hirudin is prepared by culturing a yeast transformed with an expression vector of a recombinant hirudin gene which has an expression & secretion system linking GAL1-GAL10 promoter with a secretion signal of hybridizing factor alpha A culture media particularly comprises 4% galactose, 3-6% yeast extract, and 0.2-0.8% kasamino acid. And the yeast is saccharomyces cerevisiae 2805(YEG M-HIR525)(KCTC8519P) and saccharomyces cerevisiae 2805(YEG M-HIR5)(KCTC8518P).
