<p>A novel cloning vector characterized by inverted repeated DNA sequences between which is inserted an active replicative origin. Use of the cloning vector, when introduced into a host cell, produces foreign product protein whose signal sequence is derived from other than the host cell, the signal sequence mediating membrane transport of the product protein but retaining it bound to the cell surface until released by a change in the environmental condition surrounding the host cells. The system provides a means of controlling product protein release for concentration in highly purified form.</p> |
