PURPOSE:To detect human chorionic gonadotropin (hCG) bonded to abnormal oligosaccharide, by measuring the compatibility of hCG with lectin of a plant the member of the genus Datura. CONSTITUTION:The concn. of hCG in urine to be examined is preliminarily measured by fluorescent or colorimetric immunoassay. In general, when the sugar chain terminal of hCG is sialy lated, hCG is lowered in the compatibility with Datura lectin but, in this case, since even abnormal hCG can not discriminated from normal hCG, conc. urine is desialy lated. After the conc. urine thus obtained is applied to a lectin column to be incubated, a liquid mixture of 10mM tris-hydrochloric acid buffer solution containing 0.15M sodium chloride is passed through the column. Whereupon, normal hCG rapidly flows out since has no compatibility with Datura lectin. Next, when a 0.1N acetic acid solution is replaced with said elutent to be again passed through the column, abnormal hCG rapidly flows out and, therefore, when a 4ml column is used for example, about 10ml flowing at first is fractionated by 1ml and the presence of hCG in each fraction is measured by immunoassay to make it possible to detect abnormal hCG in urine.
