PURPOSE:To efficiently obtain 2-keto-L-gulonic acid useful as a raw material for L-ascorbic acid by culturing a bacterium capable of producing 2-keto-L- gulonic acid from L-sorbose in a substrate-containing medium, recovering the product and reusing the bacterium in the next culture. CONSTITUTION:A bacterium [e.g. Pseudogluconobacter saccharoketogenes K591S (FERM-BP 1,130)] is inoculated into a liquid medium containing a substrate for producing 2-keto-L-gulonic acid, subjected to shaking culture at 28 deg.C for one day and 2-keto-L-gulonic acid produced in the culture solution is recovered. On the other hand, the bacterium is recovered, the recovered bacterium is inoculated in a new liquid medium for the next culture and the culture and the recovery are carried out at least one time so that the total reaction time is shortened and a raw material for the medium can be reduced to produce the objective 2-keto-L-gulonic acid useful as a synthetic intermediate for L-ascorbic acid industrially and advantageously. |
