PURPOSE:To exactly measure only the activity of the labeling enzyme fixed to a solid phase by bringing the unreacted labeling enzyme into contact with the undissolved matter bonded with the inhibitor of said enzyme to deactivate the labeling enzyme after subjecting said enzyme to antigen-antibody reaction. CONSTITUTION:The unreacted enzyme-labeled antibody or enzyme-labeled antigen in the antigen-antibody reaction is brought into contact with the undissolved matter bonded with the inhibitor of said enzyme to deactivate the enzyme of the enzyme-labeled antibody or enzyme-labeled antigen. The inhibitor activates the unreacted labeling enzyme by bonding only to said enzyme but maintains the activity of the solid phase to which the labeling matter is bonded. A wheat-derived inhibitor and bacterium-derived inhibitor is used for the above- mentioned inhibitor, for example, amylase. Since the unreacted labeling enzyme is removed by the undissolved matter bonded with the inhibitor, the need for a cleaning operation is eliminated and the splashing of bacteria is prevented. Only the activity of the labeling enzyme bonded to the solid phase is thus exactly measured.
