1. A method for cryoprotecting a biological specimen comprising:
chemical crosslinking of at least one polysaccharide selected from the group consisting of pullulan, dextran, dextran sulfate, scleroglucan, carboxymethyl dextran, carboxymethyl pullulan, agar, alginate, fucoidan, chitosan, hyaluronic acid, heparan-sulfate proteoglycans, heparin, heparin or heparan-sulfate mimetics, and mixtures thereof with a reticulating agent selected from the group consisting of trisodium trimetaphosphate (STMP), phosphorus oxychloride (POCl3), epichlorohydrin, formaldehydes, hydrosolubles carbodiimides and glurataldehyde under alkaline conditions at a temperature comprised between 30 and 70° C. thereby forming a hydrogel; and freezing the biological specimen in the presence of the hydrogel at a temperature below 0° C. at standard pressure conditions, and substantially free of or without dimethylsulfoxide (DMSO). |
