<p>Solid phase hybridization and competitive PCR assays for detecting and determining the concentration of target nucleic acids are disclosed. PCR target and competitor products are immobilized via a coupling agent attached to the primer, alkaline denatured, separated from sense strands and detected and quantified via hybridization with labeled probes. Alternatively, solid phase PCR is performed with target and competitor nucleic acids immobilized via the coupling agent. In a third embodiment a labeled internal primer attaches the target nucleic acid to the solid support via the coupling agent and the product of solid phase PCR is detected in situ. Kits are provided which contain reagents for performing each of the assay methods.</p> |
