<p>Isomeric cyclic (iso)adnosine-3',5'-monophosphoric acids of general formula (I) and (II) possess hypophysis-stimulating activity, in creasing thyroid hormone secretion, replacing ACTH and increasing corticosteroid prodn. Cpds. (II) are prepd. by reaction of 6-Cl-9-(ribo-beta-furanosyl-2',3'-isopropylidene)-purine with amines NHXY and phosphorylating the product with POCl3 in Me phosphate. Cpds. (I) are prepd. by cyclisation of cpds. (II) by refluxing with dicyclohexylcarbodiimide in DMF anhyd. pyridine 1 : 1 mixture. Cpds. with R' = Br are prepd. by direct bromination with aq. Br. Other R' cpds. are prepd. from the Br cpd. in known manner. (where NXY = NH2, NHMe, NMe2, NHBun, NHBut; R' = H, BR, SMe, opt. substd. NH2; R" = H or opt. substd. NH2; R = H or butyryl).</p> |
