NEW MATERIAL:Purine nucleoside phosphorylase having following physicochemical properties. Action: purine nucleoside + phosphoric acid base + ribose-1- phosphoric acid; substrate specificity: affects to nucleoside and base other than xanthosine, xanthine, cytidine, cytosine, uridine, uracil, thymine, thymidine and derivative of said compounds; optimum pH:7 to 11; stable pH: does not inactivate by treatment at about 70 deg.C for 30min at pH4.5 to 9; optimum temperature: 70 deg.C; range of suitable temperature for action: stable to treatment at 50 to 70 deg.C for 60min at pH7.0, and inactivates to treatment at 90 deg.C for 60min at pH 7; molecular weight: 113000 (gel-filtration method by TSK gel G-3000SW); molecular weight of sub-unit: 28000 (SDS electrophoresis), etc. USE:Enzyme having excellent heat resistance. PREPARATION:Microorganism such as Bacillus stearothermophilus JTS 859 is cultured. |
