The present invention relates to methods of inducing or enhancing an immune response against an immunogen in a subject. The invention further includes isolated nucleic acid vaccines, cellular vaccines, fusion proteins, expression vectors, vaccines, and immunogenic compositions for use therein.

1. An immunogenic composition comprising a DNA construct comprising:
a. a first nucleic acid sequence encoding a polypeptide having cell death inducing activity, wherein the polypeptide is perforin, and wherein the first nucleic acid sequence is operatively linked to a first promoter; and b. a second nucleic acid sequence encoding an immunogenic polypeptide, wherein the second nucleic acid sequence is operatively linked to a second promoter, and wherein the second promoter has a greater relative efficacy than the first promoter.
