PROBLEM TO BE SOLVED: To provide a matrix material suitable for amino acid sequence analysis of protein and peptides.SOLUTION: A method of specifically cleaving an N-Cα bond or a Cα-C bond in a peptide main chain includes irradiating protein or peptides with a laser beam under the existence of a compound represented by general formula described below. A reagent for specific cleavage, a hydrogen radical-releasing reagent, a matrix reagent for MALDI-ISD, a matrix for MALDI-ISD, a peptide ionization reagent for MALDI-ISD, and a kit for MALDI-ISD are also provided.
