PROBLEM TO BE SOLVED: To provide a chimeric and a humanized antibody to amyloid &bgr;, and to provide a method and a composition for therapeutic and diagnostic use in treating amyloidosis which is a group of disorders and defects associated with amyloid protein such as Alzheimer's disease.SOLUTION: The invention relates to a chimeric antibody or a fragment thereof, or a humanized antibody or a fragment thereof, which specifically binds to at least one epitope on the &bgr;-amyloid protein. The epitope comprises at least two consecutive amino acid residues predominantly involved in binding to the antibody.
