Obtaining azotophiles microorganisms IsaPro (N), comprises culturing the microorganisms in a microporous matrix substrate as in vitro culture medium comprising the number of concentrations of carbonaceous substrates (C) and nitrogen compound (N). Obtaining azotophiles microorganisms IsaPro (N), comprises culturing the microorganisms in a microporous matrix substrate as in vitro culture medium comprising a number of concentrations of carbonaceous substrates (C) and nitrogen compound (N) of which one is a priori constant throughout the inert matrix substrate including agar agar (1%), and the other gradually diffuses thus establishing a gradient of ratios C/N through a certain side distance on which IsaPro biomass (N) radially spreads until this expansion is stopped at a point dNm, or any of the substrates C and/or N become limitative. The microporous agar matrix substrate allows the diffusion of one or more of substrates C and/or N according to a diffusion coefficient De (cm 2>/sec) empirically established and that from a central well, preferably in a center of a container comprising a suitable culture medium, preferably a Petri dish containing the matrix substrate. The candidate microorganism IsaPro (N), is inoculated immediately surrounding the central well, its expansive growth that can make it radially from this point. The point of radial expansion growth of IsaPro (N), is raised at a given time, or in the case of many actinomycetes and fungi after 48 hours of incubation. The C/N ratio at this point dNm (CNd) is determined by measuring, or simply calculating, a concentration of the nutrient gradually diffusing through the matrix substrate, generally the inorganic nitrogen and rational comparison between the concentration of the inorganic nitrogen and a concentration of other nutrient such as organic carbon catabolisable by IsaPro (N).
