A method for enhancing immune response in a subject under the treatment of an infectious or malignant disease, comprising administering the subject topically, subcutaneously or intramuscularly with a pharmaceutical composition a DNA construct fused with a mammalian expression vector and a pharmaceutically acceptable carrier, wherein the DNA construct comprises (1) a polynucleotide sequence encoding Cytotoxic T-lymphocyte antigen-4 (CTLA-4) or a fragment thereof, and (2) programmed death-1 (PD-1) or a fragment thereof.
