FIELD: veterinary science; virology.SUBSTANCE: disclosed method of producing serum for diagnosis of Aleutian disease of mink (ADM) comprises obtaining high-purity viral antigen by homogenisation organs infected with ADM followed by triple freezing/defrosting of homogenate, disintegration of cell homogenate with triple ultrasonic treatment, deposition of destructed cells with double-speed centrifugation, followed by deposition of immune complexes, seal residue low-speed centrifugation, by extraction of immune complexes (IC) with minimum amounts of TOE buffer by triple extraction in cold for 1-2 hours with subsequent low-speed centrifugation, deposition of IC by high-speed centrifugation of combined extracts at 30,000 rpm for 2.5 hours, extraction of IC from sludge with minimum amounts of TOE buffer by triple extraction in cold for 1-2 hours with subsequent low-speed centrifugation, dissociation of immune complexes in an acidic medium for 1 hour in cold, separation of ADM-F from antibodies by high-speed ultracentrifugation, dissolving residue in minimum amount of carbonate buffer, ferritin separation from viral particles on a column with sepharose 6B in a carbonate buffer. Further, method includes immunising rabbits with trace amounts of purified antigen in five steps to obtain serum with high titre of antibodies to ADM - 1/16384 virus.EFFECT: obtaining diagnostic serum with high titre, making it possible to test ADM virus in various biological fluids.4 cl, 2 ex, 4 dwg |
