PROBLEM TO BE SOLVED: To provide methods, reagent compositions and kits comprising the compositions for determining the amount or activity of a luciferase being a non secretive variant of a secretive type luciferase in an experiment, where simultaneous measurement of two or more different luciferases is enabled with a single reagent capable of supporting the activity of the two or more different luciferases.SOLUTION: A method of the invention for providing higher sensitivity for a non secreted luciferase and facilitating its folding into an active conformation comprises a step of contacting a sample with an effective amount of a reagent composition comprising one or more bromides such as NaBr, a chelator, an oxidizing agent or redox buffer, and a buffer having a pH higher than 8.0; a step of contacting the sample with a substrate of the luciferase enzyme; and a step of detecting or quantifying the bioluminescence in the sample.
