Provided are: a method for measuring the yield of a cyclic nucleotide induced by a sample originating from an animal, said method comprising (a) a step for heat treating the sample originating from the animal and (b) a step for contacting a cell with the heat treated sample originating from the animal; a composition and a kit to be used therefor; and a method for reducing background in an assay system of the agonist activity of a G protein-coupled receptor existing in a sample originating from an animal with the use of a cell expressing the G protein-coupled receptor, said method comprising a step for heat treating the sample originating from the animal.
