PROBLEM TO BE SOLVED: To provide improved cytokine expression in mammalian cells by optimizing all steps of gene expression of the cytokine.SOLUTION: The invention provides a nucleic acid sequence improved for expression of IL-15 and IL-15 receptor α in mammalian cells, including codon optimization. The invention provides methods of expressing the IL-15 and IL-15Rα in a mammalian cell by transfecting the cell with the nucleic acid sequence. The invention provides pharmaceutical compositions comprising expression vectors and comprising IL-15 and IL-15 receptor α combination nucleic acid and protein used to increase stability and potency of the IL-15 and IL-15Rα in vitro and in vivo.SELECTED DRAWING: Figure 29
