The present invention relates to: a mono-PEGylated interferon-beta (IFN-β) variant; and a composition for preventing or treating hyperproliferative diseases, inflammatory diseases, autoimmune diseases, or viral infectious diseases, containing the same as an active ingredient. The mono-PEGylated IFN-β variant of the present invention has excellent anti-viral efficacy, immune-regulating function, and anti-growth function, compared with those of natural type IFN-β, and thus can be used for various hyperproliferative diseases, inflammatory diseases, autoimmune diseases, and viral infectious diseases. The present invention has an excellent pharmacologically active effect in various administration methods, and thus has a more significantly increased blood half-life than a conventional interferon preparation and has greatly expanded and improved administration paths, thereby having very excellent patient convenience.
