The present invention discloses a method of in vitro culture of wheat spikes through hybridization between wheat and maize to induce haploid embryos, wherein: the wheat spikes cut from fields or pots 24 hours after pollination with maize pollen are sprayed with 100mg 11 2,4-D and then inserted in containers with a culture solution; then the containers are placed in a climatic cabinet to culture continuously day and night; the culture solution is replaced for the first time after 3 days of culture, and then for the second time after another 3 days of culture, and then for the third time after another 4 days of culture until the culture is finished. During the course of 13-14 days from start to end of culture, when the spike culture solution is replaced each time, the volume of solution shall allow it to submerge spike stalks by 2-3 cm, and at least 2-3 cm of the stalk base is cut off with scissors to a plain end and the leaves not growing on the node of the stalk base are removed. The average rate of haploid embryo by the present invention can be stable at about 30%, with the highest rate of 70%, and the embryos develop well.
