PROBLEM TO BE SOLVED: To provide methods for detecting food poisoning bacteria specifically and simultaneously, where E. coli, salmonella, Staphylococcus aureus and Vibrio parahaemolyticus are simultaneously cultured in the same culture medium and respective target regions are detected simultaneously by PCR amplification with good amplification of the target region of Staphylococcus aureus.SOLUTION: The method of the invention comprises the steps of: growing the four bacterial species in an appropriate culture medium at the same time; extracting the genomic DNA of the four species from the obtained culture; amplifying by PCR DNA fragments comprising pyrH of E. coli, salmonella invA, dnaJ of Staphylococcus aureus, toxR of Vibrio parahaemolyticus respectively; detecting the amplification products by using electrophoresis or DNA chip at the same time, where the primer concentration in the primer set for amplifying dnaJ in the PCR solution in the amplifying step is 1.25 times or more each primer concentration in the primer sets for amplifying pyrH, invA and toxR. |
