Inactivated hog cholera vaccine is prepared by growing a vaccine virus 'by infecting a swine tissue culture monolayer e.g. swine testicle tissue with virulent hog cholera virus at a pH of 7.2 to 7.6 until substantial virus replication has occurred, freezing said resulting vaccine virus followed by thawing said vaccine virus and recovering the virulent virus, inactivating said virulent virus by (i) combining said virulent virus with aqueous toluidine blue said toluidine being blue added in an amount to provide a final concentration of 2 mcgs. of toluidine blue per ml. of vinus, and irradiating the resulting mixture with light or by (ii) use of hydroxylamine or ethylene oxide, and harvesting the inactivated vaccine (product. The harvesting of the inactivated product Imay be performed by absorbing the virus on hydrated aluminium oxide to precipitate a mixture comprising inactivated virus and hydrated aluminium oxide and subsequently reconstituting the precipitate in phosphate buffered saline.</p> |
