PURPOSE: A method for preparing GDP-fucose with high productivity and high yield is provided. CONSTITUTION: A GDP-fucose by culturing E.coli is prepared by overexpression of GDP-D-mannose-4,6-dehydratase, GDP-4-keto-6-deoxymannose-3,5-epimerase-4-reductase, phosphomannomutase, mannose-1-phosphate guanyltransferase, and guanosine-inosine kinase using recombinant E.coli. The overexpresison is controlled by an induction promoter. The culture is fed-batch culture of a pH-stat method.
