Also disclosed is the new pathway as a means for cells to down-regulate the expression of Stau1-binding mRNAs.

A method of screening for an agent that modulates SMD comprising i. incubating the agent with a stably transfected cell comprising a reporter gene with a 3′ UTR ALU element; a lncRNA comprising a ALU element that base pairs with the ALU element in the mRNA of the reporter gene; and Stau1, and ii. assaying the amount of mRNA of the reporter gene in the cell, wherein an increase or decrease in the amount of mRNA of the reporter gene relative to the amount of mRNA of the reporter gene in the absence of the agent indicates a substance that modulates SMD activity.
