EXTRACTION OF PROTEINS FROM FORMALIN-FIXED TISSUE

The tissue samples are heated at an incubation temperature between 61[deg]C and 100[deg]C for a sufficient interval to release intact proteins. This treatment continues for between 20 minutes and 16 hours. No proteases are used. The extraction buffer is a mixture of tissue protein extraction reagent (T-PER) (Pierce) and Lammli (gellade) buffer. Sodium dodecyl sulfate (SDS) detergent is included. The sample is de-paraffined before protein isolation. Further details of method and reagents are provided. An independent claim is included for a corresponding kit, used in the determination.
