The method may be used, for example for producing fermentable sugars for the production of bioethanol from lignocellulosic material. A nucleic acid molecule, comprising a polynucleotide sequence selected from the group consisting of: (a) a polynucleotide sequence encoding a polypeptide having cellobiohydrolase II activity, the polypeptide comprising the sequence consisting of amino acids 1-25 directly linked to amino acids 64-478 of SEQ ID NO:12 or the sequence consisting of amino acids 19-25 directly linked to amino acids 64-478 of SEQ ID NO:12; (b) a polynucleotide sequence encoding a polypeptide having cellobiohydrolase II activity, the polypeptide having at least 80% identity to the sequence consisting of amino acids 1-25 directly linked to amino acids 64-478 of SEQ ID NO:12 or the sequence consisting of amino acids 19-25 directly linked to amino acids 64-478 of SEQ ID NO:12; (c) a polynucleotide sequence comprising a cDNA comprising the coding sequence of the polynucleotide sequence set forth in SEQ ID NO:11; (d) a polynucleotide sequence comprising a cDNA comprising the coding sequence of the polynucleotide sequence encoding a polypeptide having cellobiohydrolase II activity contained in the E. coli strain deposited under accession number DSM 22946; (e) a polynucleotide sequence comprising a cDNA encoding the amino acid sequence consisting of the sequence of amino acids 19-478 of SEQ ID NO:12; (f) a polynucleotide sequence molecule the coding sequence of which differs from the coding sequence of a nucleic acid molecule of any one of (c) to (e) due to the degeneracy of the genetic code; and (g) a polynucleotide sequence encoding a polypeptide having cellobiohydrolase II activity, the polypeptide comprising a heterologous signal sequence linked to an amino acid sequence having at least 80% identity to a sequence consisting of amino acids 19-478 of SEQ ID NO:12.
