PROBLEM TO BE SOLVED: To provide reverse genetics using non-endogenous pol I promoters.SOLUTION: A pol I promoter not endogenous to the organisms belonging to the same taxonomic order as a host cell is used to drive expression of a transgene in the host cell. In one embodiment, the invention provides a host cell comprising one or plural expression constructs, and the expression of RNA molecules from the expression construct is regulated by a pol I promoter not endogenous to the taxonomic order of the host cell. The invention further provides a process for expressing RNA in a host cell comprising the steps of: (i) preparing an expression construct of which expression of the subject transgene is promoted by a pol I promoter derived from a first organism; (ii) introducing the expression construct of step (i) into a host cell belonging to a taxonomic order different from that of the first organism.
