The regulation of protein synthesis at the level of translation initiation plays a key role in the control of cell growth, proliferation, and apoptosis. Translation, the mRNA-directed synthesis of proteins, occurs in three distinct steps: initiation, elongation and termination. Translation initiation is a complex process in which the two ribosomal subunits and methionyl tRNA (met-tRNAi) assemble on a properly aligned mRNA to commence chain elongation at the AUG initiation codon. The interaction between the initiation factors eiF4E and eiF4G is a major component of this process. eiF4E binds the 7-methylguanosine cap structure found at the 5' ends of most messenger RNAs. |
