POLYNUCLEOTIDE ENCODING HUMAN TELOMERASE REVERSE TRANSCRIPTASE AND METHOD FOR PREPARING HUMAN TELOMERASE REVERSE TRANSCRIPTASE IN E. COLI USING THE SAME

A polynucleotide is provided to prepare a human telomerase reverse transcriptase efficiently in E. coli. And a method for preparing a human telomerase reverse transcriptase and a method for preparing an activated human telomerase reverse transcriptase are also provided. A polynucleotide includes a sequence described as SEQ ID : NO. 1. A recombinant vector includes the polynucleotide and is pET28a(+)-hTERT. A method for preparing an activated human telomerase reverse transcriptase comprises the steps of: (a) isolating and purifying human telomerase reverse transcriptase prepared by culturing a cell transformed by the recombinant vector transformed cell; (b) phosphorylating the transcriptase obtained from the step(a) using a protein kinase Calpha kinase or an Akt protein kinase; and (c) mixing the phosphorylated enzyme with RNA of SEQ ID : NO. 2 to reconstitute the enzyme.
