An engine start control system of an automatic transmission vehicle is provided to control drive of a start motor by controlling the operation of a start relay. An engine start control system of an automatic transmission vehicle includes an ignition switch(10), a transmission control unit(20), a start relay(40), and an engine control unit(30). One end of the ignition switch is connected to an output terminal of a battery. The ignition switch supplies the voltage of the battery to any contact point selected among various contact points(Lock, Off, Acc, On, ST). The engine control unit controls the operation of a start relay when the start of an engine is demanded. The transmission control unit applies a P/N signal on the position of a transmission lever to the engine control unit in CAN(Controller Area Network) communication. The engine control unit controls the operation of the start relay according to the P/N signal applied to the transmission control unit.
