<P>PROBLEM TO BE SOLVED: To provide a method for isolating the 5'-terminals of a nucleic acid originated from a biological resource in a test tube. <P>SOLUTION: This method for catching the terminals of the nucleic acid comprises (a) a process for removing the 5'-phosphate group of a non-full length RNA molecule, (b) a process for removing a capping site from a 5'-perfect RNA molecule, and adding a proper nucleic acid. The obtained modified RNA is subjected to a reverse transcription to obtain a cDNA. Thereby, the construction, tagging and sequence determination or RNA reduction of a cDNA library containing a small scale RNA are made possible. <P>COPYRIGHT: (C)2009,JPO&INPIT |
