The present invention relates to an SMAD protein activator containing pyruvate dehydrogenase kinase 4 (PDK4), a method for inducing the activation of an SMAD protein by treating cells with PDK4, and a pharmaceutical composition for preventing or treating diseases caused by abnormal SMAD protein activation, the pharmaceutical composition containing PDK4 or a PDK4 inhibitor as an active ingredient. According to the present invention, PDK4, which is known as a protein that regulates the activity of a pyruvate dehydrogenase complex (PDC) involved in glucose oxidation, can phosphorylate amino acids of serine 463 and serine 465 of SMAD protein, which is known as a signaling material on the existing bone morphogenetic protein (BMP) signaling pathway, and thus the present invention can suggest a possibility of using PDK4 as a novel SMAD activator, and can suggest a possibility of applying PDK4 or a PDK4 inhibitor as a therapeutic use for various diseases that may be caused by abnormal phosphorylation of SMAD protein. |
