<p>The invention comprises ACTH pectic compounds and purified ACTH prepared therefrom. ACTH is mixed with pectic acid, pectinic acid having a methyl ester content of up to ten per cent or one or more metal salts of such acids, such as calcium, zinc, aluminium, sodium and potassium pectates. The pectic substance may be dissolved in water or suspended in ethanol, methanol or isopropanol. The pH of the reaction mixture is adjusted to between about 2,5 and about 5 by addition of hydrochloric, acetic, tri-chloracetic or sulphuric acid or sodium or ammonium hydroxide. The mixture is allowed to stand while the precipitate settles, centrifuged and the precipitate washed acid and dried with acetone. ACTH may be obtained from the pectic compound by elution of a column of the pectic compound with a strong acid, preferably of pH less than 1,5 e.g. sulphuric, perchloric, nitric or hydrochloric acid. The acid eluate may be mixed with acetone, methyl ethyl ketone, ethanol, propanol or other precipitating agent or it may be dialysed and the non-dialysable portion lyophilized. Numerous examples of both procedures are given.</p> |
