PROBLEM TO BE SOLVED: To provide novel agents capable of mediating target-specific RNA interference or other target-specific nucleic acid modifications such as DNA methylation.SOLUTION: There is provided a synthetic, isolated, double-stranded RNA molecule. The RNA molecule is capable of target-specific nucleic acid modifications, and the double stranded RNA molecule has a first and a second RNA strand and either the first or second RNA strand is modified or blocked at the 3' end so that dsRNA processing originates at the opposing 3' end of the RNA duplex molecule.
