摘要 <p>The invention relates to a method for preparing for and carrying out the microscopic recording of image stacks of a sample (2) from various orientation angles, wherein the sample (2) is retained in a sample retainer (3) and the sample retainer (3) defines a sample coordinate system. The sample retainer (3) itself can be translated and rotated in a space spanned by a detection coordinate system. In a first embodiment, for at least two different orientation angles (alpha, alpha 2), a projection image is recorded in the detection coordinate system and a first sample volume (V) and a second sample volume (V 2) are determined in the sample coordinate system. Then the intersection volume of said two sample volumes is determined in the sample coordinate system. The parameters necessary for image stack recording are determined in the sample coordinate system for all other orientation angles on the basis of the intersection volume. Alternatively to determining an intersection volume, a sample volume (V P) can also be defined by determining start and end parameters for image stack recording for an orientation angle. The sample (2) is then positioned in the detection coordinate system accordingly and the image stack recording is carried out. A conversion between the sample coordinate system and the detection coordinate system is carried out by means of corresponding transformation matrices.</p>
