A process for the determination of the hydroxyproline content of a biological fluid comprises treating a sample of the fluid with a strongly acidic cation exchange resin which adsorbs the free and peptide-bound hydroxyproline present, hydrolysing and adsorbed peptide-bound hydroxyproline by heating, eluting the hydroxyproline and determining its content in the eluate by means of a colour reaction. The resin is preferably a 2 to 10% cross-linked sulphonated polystyrene resin particle size 20 to 200 microns, optionally applied to inert carriers particles; others are mentioned. The eluate may be analysed by conversion of the hydroxyproline to a pyrrole compound using hydrogen peroxide or the Na salt of N-chloro p-toluene sulphonamide and formation of a colour by addition of p-dimethylamino benzaldehyde. A test pack comprises a bottle containing a strongly acidic cation exchange resin, at least two tubes for the adsorption and hydrolysis, at least one bottle containing a standard solution of hydroxyproline or a known quantity of hydroxyproline for the preparation of a standard solution, and bottles containing stable reagents for the performance of a colour reaction on hydroxyproline. |
