The present invention relates to a medium for ex vivo amplification of canine natural killer cells and, more specifically, to an ex vivo culturing method of canine natural killer cells capable of amplifying ex vivo amplification efficiency of the canine natural killer cells using the medium for ex vivo amplification of the canine natural killer cells, and preparing the canine natural killer cells in which anticancer activities are enhanced. The ex vivo culturing method of canine natural killer cells comprises: a first step of isolating the canine natural killer cells from blood collected from a canine; and a second step of ex vivo culturing the canine natural killer cells by adding the same to the medium for ex vivo amplification.
