EXPRESSION CONSTRUCTS AND METHODS FOR EXPRESSING POLYPEPTIDES IN EUKARYOTIC CELLS

The invention relates to an expression construct for the expression of polypeptides in host cells using alternative splicing. The expression construct can be used for the expression of polypeptides such as antibodies, antibody fragments and bispecific antibodies by expressing the gene products required for protein expression at the ratio leading to the highest titres or the best product quality profile.

1. An expression construct comprising in a 5′ to 3′ direction:
(i) a promoter; (ii) an optional first splice donor site; (iii) a first flanking intron; (iv) a first splice acceptor site; (v) a first exon encoding a first polypeptide; (vi) an optional second splice donor site; (vii) a second flanking intron; (viii) a second splice acceptor site; and (ix) a second exon encoding a second polypeptide, (x) wherein upon entry into a host cell, transcription of the first exon results in expression of the first polypeptide and/or transcription of the second exon results in expression of the second polypeptide.
