MICROSPHERE-BASED COMPOSITION FOR PREVENTING AND/OR REVERSING NEW-ONSET AUTOIMMUNE DIABETES

PROBLEM TO BE SOLVED: To provide a microsphere-based composition for preventing and/or reversing new-onset autoimmune diabetes.SOLUTION: A method is provided that includes using an antisense approach to reverse and/or delay an autoimmune diabetes condition in vivo. Oligonucleotides are targeted and bind to primary transcripts CD40, CD80, CD86 and their combinations. In one aspect of the disclosure, three AS-oligonucleotides targeted to the CD40, CD80 and CD86 transcripts are synthesized, and an aqueous solution of the oligonucleotide mixture is prepared and mixed with an aqueous polymer solution. Microspheres containing the oligonucleotides are formed, and these are delivered to NOD mice by injection.
