The present invention relates to a composition comprising an expression or activity inhibitor of p34 as an active ingredient for treating or suppressing metastasis of cancer. According to the present invention, knock-down of p34 protein promotes nuclear localization of PTEN by causing mono-ubiquitination of PTEN to suppress Akt pathway associated with survival, proliferation, invasion, and metastasis of a tumor, thereby can significantly reduce the tumor-developing ability and the colony-forming ability of a variety of cancer cells co-expressing the PTEN and NEDD4-1. Accordingly, the expression inhibitor of p34 gene or activity inhibitor of p34 protein can be valuably used as a therapeutic agent or metastasis inhibitor of cancer.
