The present invention relates to a bio-sensing method using localized surface plasmon resonance (LSPR). The bio-sensing method using LSPR according to the present invention measures wavelengths and the intensity of lights which are reflected after simply radiating probe lights to the rear side of a plate without using a prism or an optical system when measuring the change in optical properties of LSPR, thereby stably bio-sensing as the probe lights are not penetrated through a specimen and maximizing utilization by enabling the introduction of a complex specimen control unit and equipment for other purposes.
