Provided are 2,6,9-trisubstituted purine derivatives of formula I and the preparation method and use thereof, wherein R1, R2, R3 and R4 are as defined in the description. In vitro cell experiments proved that the compounds can effectively inhibit the growth of various tumour cells, and can be developed into anticancer drugs; animal experiments proved that the compounds have a significant anti-EV71 virus effect in vivo, and can be developed into drugs for treating and preventing EV71 virus infections.
