PROBLEM TO BE SOLVED: To provide a method of using a nucleic acid-based gene silencing effector for generating tumor-free pluripotent stem cells.SOLUTION: A gene silencing effector contains the nucleic acid sequence of SEQ.ID.NO.3, and interferes in the gene expression of cellular CDK2, cyclin D, MECP1-p66, and MECP2. A method of the invention comprises a step for introducing a gene silencing effector into a non-in vivo cellular substrate. The expression of the gene silencing effector is controlled by a drug inducible gene expression promoter. The gene silencing effector can be expressed to the amount exceeding the ordinary expression amount of mir-302 in human embryonic stem cells with an inducible agent so that tumorigenic potential of cells are suppressed by interfering in CDK2, the cyclin D, MECP1-p66, and MECP2 to generate tumor-free pluripotent stem cells. |
