Rationally designed antibodies

Antibodies or fragments thereof having CDR regions replaced or fused with biologically active peptides are described. Flanking sequences may optionally be attached at one or both the carboxy-terminal and amino-terminal ends of the peptide in covalent association with adjacent framework regions. Compositions containing such antibodies or fragments thereof are useful in therapeutic and diagnostic modalities.

A method of stimulating proliferation, differentiation, or growth of promegakaryocytes or megakaryocytes, comprising contacting promegakaryocytes or megakaryocytes with an effective amount of an immunoglobulin molecule or fragment thereof comprising one or more thrombopoietin (TPO) mimetic peptides, wherein: a) said one or more TPO mimetic peptides replace at least a portion of one or more complementarity determining regions (CDRs) or b) said one or more TPO mimetic peptides are fused to one or more CDRs, wherein said immunoglobulin molecule or fragment thereof comprising the one or more TPO mimetic peptides binds to a TPO receptor, and wherein said immunoglobulin or fragment thereof comprising one or more TPO mimetic peptides comprises one or more immunoglobulin framework regions.
