PROBLEM TO BE SOLVED: To provide a processing enzyme including an N-terminally attached tag derived from a highly basic protein from thermophilic bacteria, and useful for modifying a protein.SOLUTION: There is provided a method for producing a processing enzyme useful for modifying a protein, which is selected from oxidoreductase, transferase, hydrolase, lyase, isomerase, and ligase. The processing enzyme includes a tag which facilitates high-yield expression of the soluble tagged processing enzyme, high purity after a single cation exchange chromatography purification process, and simple removal of the processing enzyme after processing a target protein precursor, can be produced with a high yield, and can be efficiently separated from a modified protein after use.
