The present invention relates to a neuroprotective composition comprising a Vitexagnus-castus extract or a fraction thereof as an active ingredient. More specifically, the Vitexagnus-castus extract or the fraction thereof of the present invention has a protective effect for brain nerve cells and a low cytotoxicity due to an inhibitory activity on virulence of glutamate for N18-RE-105 cell strain having characteristics of brain cells. Thus, the Vitexagnus-castus extract or the fraction thereof can be valuably used as a pharmaceutical composition for neuroprotection, a pharmaceutical composition for preventing and treating brain diseases, or a health food.
