The present invention relates to the production of an ElonginB/ElonginC/Vif/CBFβtetramer complex comprising full-length Vif protein. The present invention provides an assay for screening any agent that inhibits the ability of Vif to bind with Cul5. The invention provides an agent identified by the screening methods and methods of treatment using the identified agent. The invention also provides compositions that inhibit Vif-Cul5 binding based upon regions identified in Vif and Cul5 that mediate Vif-Cul5 binding.
